    Linkage and mapping analyses of the no glue egg gene Ng in the silkworm (Bombyx mori L.) using simple sequence repeats (SSR) markers

    In the silkworm, Bombyx mori, no glue egg is mainly controlled by Ng (No glue) gene, which is located on the 12th chromosome. Owning to a lack of crossing over in females, reciprocal backcrossed F1 (BC1) progenies were used for linkage analysis and mapping of the Ng gene based on the simple sequence repeats (SSR) linkage map using silkworm strains H9 and P50, which are Ng mutant and normal to egg, respectively. The Ng gene was found to be linked to three SSR markers. Using a reciprocal BC1M cross, we constructed a linkage map of 36.4 cM, with Ng mapped at 15.9 cM and the nearest SSR marker at a distance of 7.4 cM. Based on fine genome map of domesticated silkworm (B. mori), the result of Kaikoblast show that the physical distance between the near markers (containing Ng gene) is 181.7 Kb. Further analysis show that BGIBMGA005833, BGIBMGA005835 and BGIBMGA005836 are closer to Ng, and the BGIBMGA005835 is nearest to Ng, which physical distance is 44 Kb.Key words: Gene location, linkage analysis, microsatellite, Ng, silkworm

    Molecular characterisation of canine parvovirus strains circulating in China

    Canine parvovirus (CPV) was first isolated at 1978 in the USA. Analysis of CPV isolates by monoclonal antibodies and restriction enzymes have shown that after the first emergence of CPV (CPV-2) it evolved to give rise to new antigenic types, which were designated CPV types 2a, 2b and 2c. These new types have replaced the original CPV type 2, although the proportions of each of the new antigenic types vary in different countries. In China, CPV infections were first observed in 1982, however, there has been no information concerning the antigenic types of CPV prevailing in China now. In this study, we designed a PCR assay to type canine parvovirus strains in fecal samples collected from symptomatic dogs from 2006 to 2009. Our data showed that the CPV prevalent strain is mostly 2b, the proportion of CPV-2a is very low, no CPV-2c and CPV-2 were observed

    Physiological response of natural C-taklimakanensis BRPan et GMShen to unconfined groundwater in the hinterland of the Taklimakan Desert

    Calligonum. taklimakanensis B.R.Pan et G.M.Shen is an indigenous species that grows in the Taklimakan Desert. This study shows the relationship between C. taklimakanensis B.R.Pan et G.M.Shen and water conditions in the hinterland of the desert. The results show that: (1) Depth of water table is an important factor that affects water potential (Psi(p), Psi(A)), osmotic potential (Psi(sat), Psi(tlp)), relative water content (RWCtlp, ROWCtlp), and transpiration rate. (2) The degree of mineralization has a significant impact on the water potential of plants. A high degree of mineralization can strongly reduce plant productivity. (3) C. taklimakanensis B.R.Pan et G.M.Shen reduces the temperature of assimilation sticks through a high transpiration rate and maintains relatively high water content to adapt to drought and hot weather conditions in the hinterland of the desert. In addition, C. taklimakanensis B.R.Pan et G.M.Shen adapts to the water status in the desert through self-regulation or even sacrificing productivity

    Photosynthesis responses of endemic shrubs of Taklimakan Desert to adverse temperature, humidity and radiation

    Under the native habitat conditions, the seasonal gas exchange characteristics of two natural endemic plant species, Calligonum taklimakanensis B.R. Pan & GM. Shen and Tamarix taklamakanensis M.T. Liu, which are located in the hinterland of the Taklimakan Desert, are measured and compared by Li-6400 photosynthesis system. The results indicate that temperature (degrees C), solar radiation (PAR), soil water content (SWC), and other environmental factors have obvious seasonal variations and the gas exchange characteristics of two plants have different changes in different growing seasons. For C. taklimakanensis, both in July and September, its daily changes of net photosynthetic rate tend to be obvious double peak curve, but in July its peak appeared earlier. Besides its maximum net photosynthetic rate (P-max), apparent quantum efficiency (Phi), range of effective photosynthetic radiation significantly less than that in September. Moreover, its water use efficiency (WUE) in July was also lower than that in September due to the higher transpiration rate (T-r). For T. taklamakanensis, although its daily change of net photosynthetic rate is a single peak curve in September, its peak time has not changed, and except that its WUE is higher in September like C. taklimakanensis, the maximum net photosynthetic rate (P-max), apparent quantum efficiency (Phi), light saturation point, and range of effective photosynthetic radiation has not changed or slightly declined. That is to say C. taklimakanensis select a season that habitat was better (like September) to progress relative effectively photosynthesis accumulation, in contrast, T. taklamakanensis still keep a relatively stable photosynthesis rate in different growth seasons. The difference of gas exchange characteristics of the two plants in different seasons shows that adaptation strategies of the two plants to extreme conditions in desert are different. Besides, both the higher photosynthetic accumulation rate and the higher water use efficiency in September also indicate that these two endemic desert shrubs possess the abilities and strategies to make the best of limited natural resources

    Responses of yellow catfish (Pelteobagrus fulvidraco Richardson) exposed to dietary cyanobacteria and subsequent recovery

    A 120-day toxicity experiment was conducted to investigate the effect of dietary cyanobacteria on the growth and liver histopathology of yellow catfish, and subsequent recovery when the fish were free of cyanobacteria. Three experimental diets were formulated: the control (cyanobacteria-free diet), low-cyanobacteria diet (LCD, 32.3 mu g microsystins/g) and high-cyanobacteria diet (HCD, 71.96 mu g microsystins/g). Each diet was fed to fish for 60 days and then all fish were free of cyanobacteria for a further 60 days. The results showed that a significant decrease in the specific growth rate (SGR) was observed in both fish fed with the LCD and HCD after a 1st 30-day exposure period, however, no significant difference in the SGR between the LCD and control groups was observed after a 2nd 30-day exposure period. At the end of the 60 days exposure, all examined liver tissues in both doses exhibited what appeared as dose-dependent histopathological modifications. After a 60-day recovery, there were no significant differences in the SGR among groups, while no obvious histopathological alteration was observed in livers of fish previously fed with the LCD. The results indicate that the LCD-treated fish have a full recovery after a 60-day recovery, but the HCD-treated fish did not. (C) 2012 Elsevier Ltd.     Acupuncture for chronic neck pain: a pilot for a randomised controlled trial

    Background: Acupuncture is increasingly being used for many conditions including chronic neck pain. However the evidence remains inconclusive, indicating the need for further well-designed research. The aim of this study was to conduct a pilot randomised controlled parallel arm trial, to establish key features required for the design and implementation of a large-scale trial on acupuncture for chronic neck pain. Methods: Patients whose GPs had diagnosed neck pain were recruited from one general practice, and randomised to receive usual GP care only, or acupuncture ( up to 10 treatments over 3 months) as an adjunctive treatment to usual GP care. The primary outcome measure was the Northwick Park Neck Pain Questionnaire (NPQ) at 3 months. The primary analysis was to determine the sample size for the full scale study. Results: Of the 227 patients with neck pain identified from the GP database, 28 (12.3%) consenting patients were eligible to participate in the pilot and 24 (10.5%) were recruited to the trial. Ten patients were randomised to acupuncture, receiving an average of eight treatments from one of four acupuncturists, and 14 were randomised to usual GP care alone. The sample size for the full scale trial was calculated from a clinically meaningful difference of 5% on the NPQ and, from this pilot, an adjusted standard deviation of 15.3%. Assuming 90% power at the 5% significance level, a sample size of 229 would be required in each arm in a large-scale trial when allowing for a loss to follow-up rate of 14%. In order to achieve this sample, one would need to identify patients from databases of GP practices with a total population of 230,000 patients, or approximately 15 GP practices roughly equal in size to the one involved in this study (i.e. 15,694 patients). Conclusion: This pilot study has allowed a number of recommendations to be made to facilitate the design of a large-scale trial, which in turn will help to clarify the existing evidence base on acupuncture for neck pain

    Development of specific PCR assays for the detection of Cryptocaryon irritans

    Cryptocaryon irritans is one of the most important protozoan pathogens of marine fish, causing the “white spot” disease and posing a significant problem to marine aquaculture. In the present study, a C. irritans-specific reverse primer (S15) was designed based on the published sequence of the second internal transcribed spacer (ITS-2) of ribosomal DNA (rDNA) of C. irritans and used together with the conserved forward primer P1 to develop a specific polymerase chain reaction (PCR) assay for direct, rapid, and specific detection of C. irritans. The specificity of these primers was tested with both closely and distantly related ciliates (Pseudokeroronpsis rubra, Pseudokeroronpsis carnae, Euplotes sp. 1, Ichthyophthirius multifiliis, Pseudourostyla cristata, and Paramecium caudaium), and only C. irritans was detected and no product was amplified from any other ciliates examined in this study using the specific primer set P1-S15. The specific PCR assay was able to detect as low as 45 pg of C. irritans DNA and a nested PCR assay using two primer sets (P1/NC2, P1/S15) increased the sensitivity, allowing the detection of a single C. irritans. The species-specific PCR assays should provide useful tools for the diagnosis, prevention, and molecular epidemiological investigations of C. irritans infection in marine fish
